METHODS: Primarily cultured mouse pancreatic βcells were pretreated by 0.1 mmol・L-1 genistein for 12 hours. The membrane potentials of βcells were recorded by using perforated patchclamp techniques with wholecell mode. RESULTS: Genistein treatment significantly decreased the resting membrane potentials of βcells under the condition of both 2 mmol・L-1 glucose and 0.2 mmol・L-1 diazoxide, the opener of KATP channels. After stimulated by 15 mmol・L-1 glucose, normal βcells depolarized with short latent periods and provided the bursts of action potentials. In contrast, mouse βcells treated by genistein had significant increase in latent periods before depolarization, significant attenuation of action potentials, and significant elevation in membrane potentials at interval of action potentials. CONCLUSION: Longterm treatment with genistein impairs the responses of pancreatic βcells to glucose, and it is indicated that decrease in tyrosine kinases activity causes the dysfunction of pancreatic βcells. 【Keywords】 genistein; pancreatic βcells; membrane potentials 【摘要】 目的评职称论文: 观察长期降低蛋白酪氨酸激酶活性对胰岛β细胞的评职称论文影响. 方法: 原代培养的小鼠胰岛β细胞被0.1 mmol・L-1木黄酮(蛋白酪氨酸激酶抑制剂)处理12 h. 采用穿孔式全细胞记录方法记录胰岛β细胞膜电位的变化. 结果: 在2 mmol・L-1葡萄糖条件下以及0.2 mmol・L-1二氮嗪(KATP通道开放剂)的作用下,木黄酮处理组细胞的静息电位均较对照组明显升高. 在15 mmol・L-1葡萄糖的刺激下,小鼠正常胰岛β细胞经短暂的潜伏期后发生去极化,爆发动作电位,而木黄酮处理组细胞的去极化潜伏期明显延长,动作电位幅度明显减弱,动作电位间期的膜电位明显抬高. 结论: 木黄酮长期处理可损害小鼠胰岛β细胞对葡萄糖的反应,提示蛋白酪氨酸激酶活性降低可能是胰岛β细胞功能障碍的重要原因. 【关键词】 染料木黄酮;胰岛β细胞;膜电位 0引言 葡萄糖引发胰岛素分泌的过程是:在胰岛β细胞,葡萄糖代谢产生的ATP关闭ATP敏感性钾通道(KATP通道),引起细胞去极化,进而激活电压依赖性钙通道,引起Ca2+内流. 胞内Ca2+浓度升高激发胰岛素分泌[1]. 目前已知蛋白酪氨酸激酶在维持胰岛β细胞正常功能中具有重要作用[2]. 蛋白酪氨酸激酶抑制剂木黄酮急性处理胰岛β细胞可引起细胞电活动和胰岛素分泌的改变[3]. 但是,长期降低胰岛β细胞中蛋白酪氨酸激酶活性对葡萄糖引起的胰岛β细胞电活动的影响尚不清楚. 我们用蛋白酪氨酸激酶抑制剂木黄酮预处理原代培养的小鼠胰岛β细胞12 h后,记录了小鼠胰岛β细胞在静息状态下以及受葡萄糖刺激后其膜电位的变化.
