METHODS: Four breast cancer cell lines (MCF7, MDAMB231, SKBR3 and Bcap37) were incubated with different concentrations of resveratrol or/and 5FU for 48 h, then cell survival rate was measured by MTT assay and cell morphological change was observed by phase contrast microscope. Apoptosis was detected by flow cytometry (Annexin V/PI staining) analysis and Hoechst33258 staining. RESULTS: Resveratrol could inhibit the growth of MCF7, MDAMB231, SKBR3 and Bcap37 to different extents. The IC50 values of resveratrol in MCF7, MDAMB231, SKBR3 and Bcap37 cells were 65, 207, 139 and 213 μmol/L, respectively. The IC50 value of 5FU in MCF7 cells was 13 μmol/L. When 5FU and 65 μmol/L resveratrol were combined, the IC50 value of 5FU in MCF7 cells was decreased to 9 μmol/L. Compared with the treatment with 13 μmol/L 5FU or 65 μmol/L resveratrol alone, the percentage of Annexin V(+)/PI(-) MCF7 cells increased when these 2 drugs were used in combination. The number of apoptotic MCF7 cells calculated with flow cytometry was accordant with the result got with Hoechst33258 staining. CONCLUSION: Resveratrol interacts synergistically with 5FU to induce the apoptosis of MCF7 cells, which suggests that resveratrol is a secondary chemotherapeutic medicine for treating breast cancer. 【Keywords】 resveratrol; chemotherapy; apoptosis; breast neoplasms 【摘要】 目的小论文下载:研究白藜芦醇联合5FU促进人乳腺癌细胞MCF7凋亡的小论文下载作用. 方法:4种人乳腺癌细胞系(MCF7,MDAMB231,SKBR3和Bcap37)与不同浓度的白藜芦醇或/和5FU共孵育48 h,MTT法检测细胞存活率,并用相差显微镜观察细胞形态学改变. 用流式细胞术(检测细胞凋亡相关指标Annexin V/PI)和Hoechst33258染色检测细胞凋亡. 结果:白藜芦醇能够不同程度地抑制4种人乳腺癌细胞系(MCF7, MDAMB231,SKBR3和Bcap37)的生长. 白藜芦醇对MCF7,MDAMB231,SKBR3和Bcap37细胞的IC50(半数抑制浓度)分别为65,207,139和213 μmol/L. 单用5FU对MCF7细胞的IC50为13 μmol/L,联合使用5FU和白藜芦醇的IC50分别为9 μmol/L和3 μmol/L. 与单用组相比,65 μmol/L白藜芦醇和13 μmol/L 5FU联合使用后,MCF7细胞Annexin V水平升高. Hoechst33258荧光染色和流式细胞术分析MCF7细胞的结果一致. 结论:白藜芦醇与5FU合用可协同促进MCF7细胞凋亡,提示白藜芦醇可能用作治疗乳腺癌的二线化疗药. 【关键词】 白藜芦醇;化疗;细胞凋亡;乳腺癌 0引言 中药植物的提取物与化疗药物的合理配伍,可大大提高肿瘤治疗的疗效. 白藜芦醇(resveratrol)化学名称为芪三酚,化学结构为3,5,4’三羟基二苯乙烯(3,5,4’_trihydroxystilbene),分子式C14H12O3,Mr 128.25,属非黄酮类多酚化合物,白藜芦醇可拮抗消化、血液、呼吸、生殖等多个系统来源的多种肿瘤细胞的增殖[1-4],对肿瘤细胞的发生、促进、发展3个阶段均有抑制作用[1]. 本研究将白藜芦醇与临床常用化疗药5FU联合应用,观察其对5FU在促进细胞凋亡中的增效作用. 1材料和方法 1.1材料四种人乳腺癌细胞系MCF7,MDAMB231,SKBR3和Bcap37均为本室保存细胞;白藜芦醇(纯度大于99%,溶于500 g/L的丙二醇)由西安绿康天然药物研究所提供,用时用含10%小牛血清的RPMI 1640细胞培养液稀释至工作浓度;5FU购于第四军医大学附属一院西药房;RPMI 1640培养基为美国Gibco公司产品;小牛血清购自杭州四季青生物材料工程有限公司;四氮唑溴盐(MTT)、Annexin V/PI双标试剂盒和Hoechst33258购自美国Sigma公司;其余试剂均为国产分析纯. BX60荧光显微镜和倒置相差显微镜及照像系统为日本Olympus产品;遥控酶标分析仪为奥地利TECAN公司产品;FACSCalibur流式细胞仪为美国BD公司产品.
