Assessment of pharmacokinetics and bioequivalence of amlodipine besylate in healthy volunteers
【Abstract】 AIM: To study the pharmacokinetics parameters and bioavailability of amlodipine besylate tablets. METHODS: An open, randomized and twoperiod crossover study with a twoweek washout interval was performed in 18 healthy volunteers. Concentrations of amlodipine besylate in plasma were assayed by liquid chromatographicmassmass spectrometric method after a single oral dose of 10 mg of the tested or the referenced tablets. The pharmacokinetics parameters, AUC, Tmax and Cmax were analyzed by two oneside t test and (12α) 90%. RESULTS: The main pharmacokinetics of the tested and referenced tablets were: Tmax: (8.9±2.5) and (8.2±2.2) h; Cmax: (5.1±2.3) and (5.0±2.2) g/L; T1/2: (45±10) and (48±24) h; AUC（0-120 h）: (191±69) and (196±53) μg/（L・h）. The relative bioavailiability of amlodipine besylate was (99±23)%. CONCLUSION: The results of the statistical analysis show that the two formulations are bioequivalent.

【摘要】 目的妇产科医学论文： 研究苯磺酸氨氯地平片在健康人体的妇产科医学论文药代动力学特征及其生物等效性评价. 方法： 18名健康志愿受试者，口服苯磺酸氨氯地平试验品和参比品10 mg，用标准二阶段交叉设计自身对照试验方法，以液相色谱质谱质谱联用法测定服药后120 h内不同时刻的血药浓度，计算主要药动学参数，并采用方差分析和双单侧t检验（12α）90%置信区间进行生物等效性评价. 结果： 受试者分别口服受试品和参比品后，其主要药动学参数Tmax, Cmax, t1/2和AUC（0-120 h）分别为： （8.9±2.5）和（8.2±2.2） h；（5.1±2.3）和（5.0±2.2） μg/L；（45±10）和（48±24） h；（191±69）和（196±53） μg/(L・h). 受试品的相对生物利用度为 (99±23)%. 结论： 结果表明，两种制剂具有生物等效性.

0引言
苯磺酸氨氯地平片（Amlodipine besylate），临床上主要用于高血压和心绞痛的治疗［1］. 与同类型的其他药物相比，该药有服药量低、药效维持时间长等特点. 由于氨氯地平的血药浓度低，我们采用精确度和灵敏度更高的液相质谱联用法测定血浆中氨氯地平浓度.为确保用药质量，我们评价苯磺酸氨氯地平在人体内的生物效应，并对苯磺酸氨氯地平片和络活喜片在健康志愿者体内的药物代谢动力学和生物等效度进行了比较，旨在为临床合理用药［2-5］提供理论依据.
1材料和方法
1.1材料
1.1.1仪器TSQ型液相色谱质谱质谱联用仪，配有大气压化学电离源（APCI源）以及Xcalibur 1.4数据处理系统，Surveyor输液泵（美国Finnigan公司）. 色谱柱ZorbaxXBDC18柱，5 μm粒径，150×4.6 mmI.D（美国Agilent公司）.
