| | 妇产科医学论文 作者:柳艳,李磊,刘王莹,陈茂勇,吴倩 【关键词】 丹酚酸;抗氧化;DNA损伤;化学发光 摘要: 目的妇产科医学论文 探讨丹酚酸抗氧化活性及其对DNA氧化损伤的妇产科医学论文保护作用机制。方法 运用自由基反应模型观察丹酚酸对二苯代苦味酰肼自由基(DPPH)和羟自由基的淬灭效应,使用邻菲罗啉(Phen)-CuSO4-VC-H2O2-DNA化学发光体系测定丹酚酸对DNA损伤的抑制作用。高效液相色谱(HPLC)测定丹酚酸的组成。结果 丹酚酸水提物(AE)、乙酸乙酯萃取物(EE)、大孔吸附树脂纯化物(RE)和对照维生素(VC)、二丁基羟基甲苯(BHT)对DPPH自由基的半数清除浓度(IC50)分别为1412,153,146,158和715μg/ml。化学发光体系中,丹酚酸的加入能明显抑制并延迟DNA损伤。AE、EE、RE抑制发光50%浓度(IC50)值分别为12936,620,492μg/ml。丹酚酸清除DPPH自由基能力和对羟自由基攻击的DNA损伤保护作用受丹酚酸组成和丹酚酸B含量的影响。结论 丹酚酸属于预防型和断链型抗氧化剂,并可有效清除DPPH和羟自由基,对DNA损伤有明显保护作用。丹酚酸的抗氧化活性及对DNA损伤的保护作用与丹酚酸B含量有关。 关键词: 丹酚酸;抗氧化;DNA损伤;化学发光 Antioxidant activiy and protection effect of salvianolic acids on DNA damage Abstract: Objective To study on the antioxidant activity of salvianolic acids and mechanisms of its protection against DNA antioxidative damage.Methods Free radical model was employed to investigate the scavenging activity to 1,1-diphenyl-2-picryl-hydrazyl(DPPH) and hydroxyl free radicals.The mechanisms of the effects of salvianolic acids on protection against DNA damage was measured by chemiluminescence on the reaction system of phenanthroline (Phen)-CuSO4-VC-H2O2-DNA.Salvianolic acids composition in samples were determined according to HPLC analysis.Results The hemi-inhibitable concentration(IC50) of crude aqueous extract(AE),ethylacetate extract(EE),resin extract(RE),ascorbate(VC) and butylated hydroxytoluene(BHT) to DPPH free radicals were 141.2,15.3,14.6,15.8 and 71.5μg/ml.Three extracts could obviously inhibit and delay DNA damage caused by hydroxyl free radicals.The IC50 valvue of chemiluminescence of AE,EE and RE were 1293.6,62.0,and 49.2μg/ml.The scavenging activity and protective effects against DNA damage were effected by salvianolic acids composition and salvianolic acid B level.Conclusion Salvianic acids can effectively scavenge DPPH and hydroxvl free radicals,protect against DNA damage which belongs to preventive and chain-breaking antioxidants,and the effects are related to salvianolic acid B level. Key words: salvianolic acid;antioxidant;DNA damage;chemiluminescence 丹参是我国防治心脑血管病的传统药物,对冠心病、高血压、糖尿病肾病及癌症等疗效确切。近20多年来的药效研究表明,丹酚酸是丹参活血化瘀作用的主要功能成分〔1〕。自由基参与机体多种疾病的发生和发展过程,清除自由基、提高机体抗氧化能力,是许多药物防治慢性疾病的重要途径〔2,3〕。从化学结构上分析,丹酚酸类化合物是酚羟基的供体,具有抗氧化活性的结构基础。本文研究了丹酚酸对二苯代苦味酰肼自由基(DPPH)的清除效应,并探讨其对体外DNA氧化损伤的保护作用及可能机制,为进一步研究丹酚酸药理作用提供科学依据。
