摘要： 目的医学社会实践论文 探讨天麻素对染铅大鼠脑细胞凋亡及学习记忆功能的医学社会实践论文影响。方法 选择健康雄性Wistar大鼠50只，随机分为5组，分别为实验1，2，3，4组和对照组。除对照组外，各组大鼠均给予02%醋酸铅水溶液自由饮用，实验2，3，4组给予天麻素溶液灌胃，浓度分别为100，200，400ng/kg。用Y型迷宫测试学习记忆，用流式细胞仪测定脑细胞凋亡率。结果 （1）在Y型迷宫试验中，实验1组大鼠受电击次数显著高于实验2，3，4组和对照组（P<0.01);大鼠学习受电击次数与天麻剂量呈明显负相关。(2)实验1组脑细胞(包括海马、小脑和大脑皮层)凋亡率显著高于实验2，3，4组和对照组（P<001）；大鼠脑细胞凋亡率与天麻素剂量呈明显负相关。结论 天麻素对染铅大鼠脑细胞凋亡和学习记忆功能损害具有改善作用。

Effect of gastrodin on apoptosis of brain cells of rats induced by lead
Abstract： Objective To explore the effect of gastrodin on the apoptosis of brain cells and the impairment of learning and memory induced by lead in rats.Methods 50 male Wistar rats were chosen and random divided into 5 groups,respectively;1,2,3,4and control,10 rats in every group.Except for control all rats were given solution of 0.2%lead acetate,and at the same time group 2,3 and 4 were give gastrodin at the dosage of 100,200 and 400mg/kg respecitively.The ability of learining and memory of each group was measured by Y-maze test,The apoptosis of brain cells was determinged by flow cytometry(FCM).Results (1)The number of electric shocks of group1 was significantly increased compared with 2,3,4and control in Y-maze test (P<0.01);and there was a significant dose-response relationship between electric shocks and gastrodin.(2)the apoptosis of brain cells(P<0.01);and a signficant dose-response relationship between apoptosis and gastrodin was found.Conclusion Gastrodin may improve the apoptosis of brain cells and the impairment of learning and memory induced by lead.

铅是自然界广泛存在的一种古老的重金属毒物。1991年美国疾病预防控制中心(CDC)规定儿童血铅上限值为048μmol/L。天麻(Rhizoma Gastrodiae,RG)有多种药理作用,如镇静催眠、抗惊厥、保护神经纤维、增强机体抗缺血缺氧及抗氧化能力等作用〔1〕。为了解天麻素对铅所致脑细胞凋亡的影响,本文通过Y型迷宫试验,脑细胞凋亡,研究天麻素对铅所致脑及学习记忆功能损害的改善作用。
1 材料与方法
11 材料
111 试验动物 初成年雄性Wistar大鼠(1505±694)g(山东大学实验动物中心)。
112 试验药品 醋酸铅(天津市大茂化学试剂厂);天麻素(Gastrodin)(昆明制药股份有限公司)；碘化丙啶(PI)(美国Sigma分司)。
113 试验仪器 (1)Y型迷宫每大小为15cm×15cm×30cm,底部间隔05cm铺直径2mm的铜栅;任选一臂为安全区,在其上方安装指示灯;另两臂轮流做起步区,通以60V直流电。(2)FACASGalibur型流式细胞仪(flow cytometry,FCM)(美国Bicton-DiGkingson公司)。(3)GFA-4B型石墨炉(日本岛津公司)。
