| | 医学社会实践论文【关键词】 镉;螯合剂;睾丸毒性;解毒作用 摘要: 目的医学社会实践论文 探讨新型螯合剂N-对羟甲苯甲基-D-葡萄糖二硫代氨基甲酸钠(HBGD)和N-苯甲基-D-葡糖二硫代氨基甲酸钠(BGD)对镉致小鼠睾丸毒性的医学社会实践论文解毒作用。方法 研究镉染毒组小鼠和HBGD、BGD、二乙基二硫代氨基甲酸钠(DDTC)、二巯基丙醇(BAL)、乙二胺四乙酸(EDTA)等5种螯合剂治疗组小鼠睾丸中镉及钙、铁、锌等必需金属浓度、小鼠睾丸脂质过氧化物(LPO)值、小鼠血清乳酸脱氢酶(LDH)活性及雄性小鼠生殖能力的变化。结果 镉染毒(25mg/kg,腹腔注射)后,小鼠睾丸中镉浓度显著升高,睾丸钙、铁、锌等金属浓度、睾丸LPO值及小鼠血清LDH活性显著高于对照组。分别于染镉30min和24h后腹腔注射各螯合剂(400μmol/kg)。治疗24h后,HBGD和BGD显著抑制了上述各指标的升高(P<005),并对镉引起的雄性小鼠生育能力的下降有明显改善作用。结论 HBGD和BGD对镉致小鼠睾丸毒性有较好的解毒作用,且自身毒性小,有望成为镉的解毒剂。 关键词: 镉;螯合剂;睾丸毒性;解毒作用 Effects of new chelating agents on testicular toxicity in mice induced by acute exposure to cadmium Abstract: Objective To study the effects of new chelating agents N-p-hydroxy-methyl-benzyl-D-glucamine dithiocarbamate(HBGD)and N-benzyl-D-glucamine dithiocarbamate(BGD) on testicular toxicity in mice induced by acute exposure to cadmium.Methods HBGD,BGD,diethyldithiocarbamate(DDTC),2,3-dimercaptopropanol(BAL),and ethylene diaminetetraacetic acid(EDTA) were studied for their effects on the concentrations of cadmium(Cd),calcium(Ca),iron(Fe),and zinc(Zn) in testes,lipid peroxidation(LPO) value of testes,activity of lacatate dehydrogenase(LDH) in serum and reproductive ability of male mice induced by acute exposure to Cd.Results Mice were injected intraperitoneally with CdCl2(2.5mgCd/kg) and after 30min or 24h,they were injected intraperitoneally with chelating agents(400μmol/kg).Cd injection increased in the concentrations of Ca,Cd,Fe,Zn,and LPO value in testes as well as the activity of LDH in serum of mice.HBGD and BGD effectively prevented the increase in above indices,and clearly improved the reproductive ability weakened by exposure to Cd.Conclusion HBGD and BGD are not only more effective detoxificationon testicular toxicity in mice induced by acute exposure to Cd,but also less toxic,so they are pretect antidote for Cd. Key words: cadmium;chelating agents;testicular toxicity;detoxification 镉是一种对生物体多种脏器都有毒性的有毒重金属〔1-3〕,被机体吸收后,自然排泄非常缓慢,因此,用解毒剂驱排体内蓄积的镉是防治镉中毒的根本措施之一。但目前临床上使用的镉解毒剂由于毒副作用较大,使用效果不理想。本研究通过比较新合成的2种螯合剂N-对羟甲苯甲基-D-葡萄糖二硫代氨基甲酸钠(HBGD)与N-苯甲基-D-葡萄糖二硫代氨基甲酸钠(BGD)及现有螯合剂二乙基二硫代氨基甲酸钠(DDTC)、二巯基丙醇(BAL)及乙二胺四乙酸(EDTA)对镉致小鼠睾丸毒性的解毒作用,为开发解镉中毒的高效低毒的新型解毒剂提供依据。
