| | 论文下载 作者:朱智勇,丁晓航,朱函坪,李岩金,陈学奎,沈吉友,张涛,何培江,姚苹苹,徐芳,翁景清,龚华岳,郭志军,苏波,孙淑滨,梁伟峰 【关键词】 禽流感疫苗;免疫动物;中和抗体;效力试验 摘要: 目的论文下载 将用禽流感毒种R1203株按3种生产工艺生产的论文下载疫苗进行动物接种,观察疫苗的免疫效果。方法 用不同剂量的3种工艺生产的禽流感疫苗,分别免疫大鼠和家兔,肌肉接种2针(隔14d),首针后14和28d静脉采血,测定接种1针和2针后,动物血清中血凝抑制抗体和中和抗体的产生情况。结果 大鼠的抗体反应较家兔敏感;大鼠和家兔在疫苗接种1针后14d,都能产生血抑抗体,滴度可达1∶40,但中和抗体滴度都很低,几乎测不到;接种2针后,血抑抗体或中和抗体反应都显著高于第1针;裂解-2苗接种大鼠和家兔后的抗体反应普遍高于另2种疫苗,且家兔量-效反应明显。结论 裂解-2疫苗优于其他2种;中和抗体能正确反应疫苗的质量;H5N1禽流感疫苗的效力试验可用于免疫家兔测定中和抗体和临床前研究的动物试验,用15μg血凝素(HA)接种家兔2针,血清中和抗体滴度达1:40或以上。 关键词: 禽流感疫苗;免疫动物;中和抗体;效力试验 Observation on effect of avian influenza (H5N1) vaccine with immunization animals Abstract: Objective The vaccines produced according to the three techniques with the virus seeds R1203 of H5N1 avian influenza were used to immunize animals and to observe the effect.Methods Rats and rabbits were immunized with the different dose of the three vaccines of avian influenza (H5N1) by an intramuscular injection twice (interval 14 days). Blood was collected intravenously in the day of 14 and 28. The hemagglutination inhibition antibody and neutralization antibody occurred in the serum of the animals after the first and second inoculation.Results The antibody reaction in the rats was more sensitive than the rabbits. The rats and rabbits both could produce hemagglutination inhibition antibody in 14th day with the titer of the antibody reaching at 1:40 while the titer of the neutralization antibody was very low and hardly detected. The second inoculation were significantly higher than the first inoculation. The antibody reaction after inoculation of the split2 vaccine in the rats and rabbits were significantly stronger than that inoculated with another two vaccines. The quantumeffect reaction was distinct in the rabbits. Conclusion The production technique of split2 vaccine was better than the other two. The neutralization antibody can accurately reflect the quality of vaccines. The efficiency test should be added in vaccines research and production with the method of immune rabbits. It is suggested to immunize rabbits twice with 12μg HA with the titer of the seraneutralization antibody reaching at 1:40 or over in animal test before clinical trial. Key words: avian influenza vaccine;immune animal;neutralization antibody;efficiency test 前期禽流感疫苗毒种的检定及毒种库的建立研究结果表明,引进的2株H5N1禽流感毒种R1194和R1203均可用于疫苗的生产。最初用R1194株,按2005年《中国药典》三部制造流感全病毒疫苗的规程〔1〕,生产一批全病毒疫苗,进行接种动物的预备试验取得一定效果。2005年底,国内外禽流感疫苗研究有了新的进展,特别是在疫苗中加入佐剂,使有效接种剂量不断降低。又根据禽流感疫苗生产的工艺设计及检定研究结果,设计3种生产疫苗的纯化工艺,用R1194株或R1203株都能生产出高质量的疫苗,但R1203株生产疫苗的产量较R1194株高,因此,在正式试验时我们确定用R1203毒种生产的3种疫苗,进行动物接种,观察它们的免疫效果。结果报告如下。
