| | 全科医学论文【摘要】 目的全科医学论文 探讨褪黑素对癫痫发作中细胞凋亡的全科医学论文影响。方法 将24只成年Wistar大鼠(180~230g)随机分为NC组、PTZ组和MT组;PTZ组和MT组腹腔注射PTZ(60mg/kg),诱导癫痫发作,NC组腹腔注射等量生理盐水。MT组按体重10mg/kg于注射PTZ之前20min、立即、后1h、后2h分别经腹腔注射MT并观察1h,PTZ组于相同时间注射生理盐水。然后处死大鼠取脑,应用免疫组织化学的方法检测海马神经元Bax和Bcl-2蛋白的表达。结果 大鼠海马神经元Bax的改变:PTZ组明显高于NC组,MT组明显低于PTZ组,但高于NC组:大鼠海马神经元Bcl-2的改变:PTZ组明显高于NC组,MT组明显高于PTZ组,也高于NC组。结论 MT可以通过增强海马神经元Bcl-2的表达和降低Bax的表达而对抗癫痫中细胞的凋亡。 【关键词】 褪黑素;癫痫;戊四氮;Bax;Bcl-2 【Abstract】 Objective To explore the effect of melatonin on bcl-2 and bax of hippocampus in seizures rats induced by PTZ.Methods Twenty-four adults Wistar rats (180~230g) were randomly divided into three groups,NC group,PTZ group and MT group.A dose of PTZ(60mg/kg)was intraperitoneally injected into the rats of PTZ group and MT group and seizures was induced.The rats of NC group were administered with the same dose of normal saline.The rats of MT group were administered intraperitoneally with MT(10mg/kg)in 20 minute before injecting PTZ,at one time,1 and 2 hour after injection.Simultaneously the same dose of noraml saline was administered intraperitoneally on PTZ group.The rats were killed.To study the influence of MT on Bcl-2 and Bax immunoreactivity of neruon on hippocampus by using immunohistochemistry method.Results The number and optical density of Bax in hippocampus of PTZ group rats was higher than that of control group rats.The number and optical density of Bax in hippocampus of MT group rats was lower than that of PTZ group rats,was higher than that of control group rats;The number and optical density of Bcl-2 in hippocampus of PTZ gorup rats was higher than that of control group rats;The number and optical density of Bcl-2 in hippocampus of MT group rats was higher than that of PTZ group rats,was higher than that of control group rats.Conclusion Melatonin can resistant the apoptosis of hippocampus neruon by increased the expression of Bcl-2 and decreased the expression of Bax. 【Key words】 melatonin;seizure;pentylenetetrazole;bax;bcl-2 难治性癫痫是神经系统的常见病,发病机制至今没有完全阐明,癫痫长期发作可能造成患者的智力和认知发生改变,给社会和家庭带来沉重的负担。大量动物实验发现癫痫发作后中枢神经系统存在细胞凋亡,临床癫痫病人也大多有海马硬化存在。褪黑素(Melatonin,MT)是松果体分泌的神经内分泌激素,它对人体的作用非常广泛,近年研究发现它具有抗癫痫发作的报道[1],但是它是否对癫痫发作后神经细胞的凋亡具有保护作用研究较少,本实验旨在通过观察MT对戊四氮(pentylenetetrazole,PTZ)点燃大鼠海马神经元的Bcl-2和Bax的影响,借以了解其对神经元细胞凋亡的保护作用,弄清它在治疗癫痫中的进一步作用机制。 1 材料与方法 1.1 材料 戊四氮及多聚甲醛均为Sigma公司产品,兔抗Bcl-2和Bax抗体购自北京中山试剂有限公司,Wistar大鼠由华中科技大学同济医学院动物中心提供,羊抗兔抗体和DAB染色剂购自武汉博士德公司,褪黑素购自Alexis公司。
