| | 药学科研论文 作者:王玉玺,王 丽,王俊志,王雅贤,王惠国,李志鸿 【摘要】 目的药学科研论文 观察祛毒胶囊对LPS诱导的药学科研论文狼疮样模型小鼠几项免疫指标和病理改变的影响,探讨祛毒胶囊治疗系统性红斑狼疮(SLE)的可能机制。方法 用ELISA试剂盒检测血清中白细胞介素-6(IL-6)、γ-干扰素(IFN-γ)、肿瘤坏死因子(TNF-α)水平,对肝肾组织常规HE染色。结果 (1)血清中IL-6、IFN-γ及TNF-α水平各治疗组与模型组比较,差异有显著性(P<0.05);(2)病理结果显示模型鼠肝肾炎性改变,治疗组炎性改变减轻。结论 祛毒胶囊对狼疮样小鼠血清中IL-6、IFN-γ及TNF-α的分泌有抑制作用,能明显改善其病理状态。 【关键词】 祛毒胶囊;系统性红斑狼疮;白细胞介素-6;γ-干扰素;肿瘤坏死因子-α 【Abstract】 Objective To investigate the effect of Qudu capsule on IL-6,IFN-γ and TNF-α secretion and pathologic change in revulsive lupoid model mice with LPS and the possible mechanism of Qudu capsule in treating systemic lupus erythematosus(SLE).Methods IL-6,IFN-γ and TNF-α in serum of the model mice were detected by enzyme linked immunosorbent assay(ELISA).Hepatic and renal tissue was stained by HE.Results (1)In the treated groups,as compared with the model group,levels of serum IL-6,IFN-γ and TNF-α were significantly lower(P<0.05);(2)Pathologic results showed that liver and kidney appeared inflammation in model mice. Inflammation relieved in the treated groups.Conclusion Qudu capsule could inhibit the secretion of IL-6,IFN-γ and TNF-α and improve the pathologic lesion in model mice. 【Key words】 Qudu capsule;systemic lupus erythematosus;IL-6;IFN-γ;TNF-α 系统性红斑狼疮(SLE)是以多克隆B细胞异常活化,免疫球蛋白增多和多种自身抗体的生成及细胞内外免疫反应异常为特征的自身免疫性疾病,其病因和发病机制尚未完全清楚。研究发现SLE中存在着T细胞亚群的异常和细胞因子的异常,网络性细胞因子失衡在SLE发病中起着重要作用。现代医学多采用肾上腺糖皮质激素、免疫抑制剂、非甾体抗炎药及抗疟药治疗,但毒副作用明显,停药后易反复,故中医中药治疗该病得到广泛关注。祛毒胶囊是笔者数十年来治疗SLE行之有效的复方中成药,本研究旨在通过对狼疮小鼠血清中细胞因子的测定来探讨中药治疗SLE的免疫机制。 1 材料与方法 1.1 药品与试剂 祛毒胶囊:秦艽、紫草、生黄芪、白花蛇舌草、雷公藤、丹参、生地等,由我院药局提供;强的松片剂;大肠杆菌脂多糖(LPS);IFN-γ,IL-6,TNF-α试剂盒购于TPI Inc.17037 the 34th Ave,Suite G Lynnwood,Washington 98003 U.S.A。
