METHODS: Establish chronic renal interstitial fibrosis model in rats by peritoneal injection with aristolochic acid (AA). The total proteins from AAN models and normal rat renal tissues were separated by 2D PAGE. The differentially expressed proteins in 2DPAGE maps were analyzed using image analysis software. RESULTS: The rat models of AAinduced chronic renal interstitial fibrosis were established successfully. Clear, wellresolved and reproducible 2D PAGE patterns of AAN models and normal rat renal tissues were obtained and many differentially expressed proteins were found. CONCLUSION: The differentially expressed proteins of renal tissues can be induced by AA in Wistar rats. These proteins maybe mediate the toxicity of aristolochic acid to renal tissues. 【Keywords】 aristolochic acid; rats; renal tissues; 2D PAGE; proteome 【摘要】 目的药学毕业论文: 制备慢性马兜铃酸肾病的药学毕业论文大鼠模型,建立马兜铃酸肾病大鼠肾组织和正常大鼠肾组织的双向电泳图谱,从而展示差异表达的蛋白质,为进一步筛选介导马兜铃酸毒性作用的蛋白质分子奠定基础. 方法:应用马兜铃酸腹腔注射制备大鼠慢性马兜铃酸肾病模型,提取其肾组织中总蛋白质,采用双向聚丙烯酰胺凝胶电泳技术,对模型组和正常组肾组织中蛋白质进行差异展示. 结果:成功制备了慢性马兜铃酸肾病大鼠模型,并建立了模型组和正常组大鼠肾组织的双向电泳图谱,发现了差异蛋白质. 结论:马兜铃酸导致大鼠肾组织中蛋白质差异表达,这些差异表达的蛋白质可能介导了马兜铃酸的毒性作用. 【关键词】 马兜铃酸;大鼠;肾组织;双向聚丙烯酰胺凝胶电泳;蛋白质组 【中图号】 R587.1 0引言 自Vanhrweghem等[1]报道了因减肥药引起“中草药肾病(Chinese herbs nephropathy,CHN)”以来,含马兜铃酸成分的中草药所致的肾损害马兜铃酸肾病(aristolochic acid nephropathy,AAN)越来越受到了国内外学者的广泛重视. 由于一些常用中草药及中成药都含有马兜铃酸,因而由马兜铃酸引起的肾损害在临床上十分常见[2]. 急性AAN主要表现为急性肾小管坏死,慢性AAN表现为寡细胞性肾间质纤维化[2-3],病情多为不可逆性,而且即使停服药物,其肾损害仍然继续进展[4-5]. 绝大多数患者很快或逐渐进入终末期肾衰,需要肾脏替代治疗. 因此阐明马兜铃酸肾损害的分子机制,以进行早期的诊断和治疗(在蛋白质或基因水平进行干预和阻断)将是缓解马兜铃酸肾损害的关键. 为此,我们建立慢性马兜铃酸肾病模型,用双向聚丙烯酰胺凝胶电泳筛选和比较马兜铃酸诱导的差异蛋白质分子,为马兜铃酸导致肾损害的分子靶点研究奠定基础.
