METHODS: MI was induced by occluding the left anterior descending artery (LAD) in SD rats. MSCs were isolated from the rats, purified, proliferated and they (1 mL) were transfused through caudal vein into transplantation groups (n=10/group) at 1 d, 1, 2, and 4 weeks after MI; the same culture medium (1 mL) was transfused into the control group (n=10). Four weeks later, the heart function and myocardial ischemic size and infarct size were evaluated. RESULTS: The heart function and myocardial ischemic size and infarct size of 1day group after MI had no difference from those of the control group (P>0.05) . The improvement of heart function in 1, 2, and 4week groups were more distinct than that of 1day group (P<0.05); myocardial ischemic size and infarct size were also smaller than those of 1day group (P<0.05). Among the transplantation groups, the treatment effect of 2weeks group was the best. CONCLUSION: Subacute stage (1-2 weeks after MI) might be the optimal time for MSCs transplantation. 【Keywords】 born marrow mesenchymal stem cells;myocardial infarction;intravenous transplantation; heart function 【摘要】 目的药学论文范文: 探讨心肌梗死后骨髓间充质干细胞(MSCs)移植的药学论文范文最佳时间. 方法: 结扎大鼠左冠状动脉制备心肌梗死模型,将分离培养的大鼠MSCs经尾静脉分别移植到1 d, 1, 2, 4 wk移植组体内(各组n=10),对照组则注入等量培养液(n=10). 4 wk后测定左心功能指标和心肌缺血梗死面积. 结果: 心肌梗死后1 d移植组左心功能和心肌缺血梗死面积与对照组无明显差别(P>0.05),而1, 2, 4 wk移植组与1 d移植组对比左心功能明显改善(P<0.05),心肌缺血梗死面积显著减小(P<0.05),其中2 wk移植组效果优于1 wk移植组和4 wk移植组(P<0.05). 结论:心肌梗死后1~2 wk亚急性期进行细胞移植效果最好. 【关键词】 骨髓间充质干细胞;心肌梗死;静脉移植;心功能 0引言 传统观点认为心肌细胞属于不可再生细胞,一旦损害将无法修复,死亡的心肌细胞被成纤维细胞所代替,引起心脏结构的改变,心力衰竭的发生. 现有的内外科治疗手段并不能很好地解决功能心肌细胞的丧失问题. 随着国内外干细胞试验研究的深入,干细胞移植治疗心肌梗死以及晚期心功能不全正日益成为一种新的治疗方法[1-2]. 骨髓间充质干细胞(MSCs)由于能够自我复制,具有分化的多功能性,并且无免疫排斥反应,因而在临床中具有广泛的应用前景[3-4]. 但是骨髓间充质干细胞移植在什么时候进行才能使机体获益最大?这个问题一直是细胞移植领域备受关注的课题. 本实验我们探讨在心肌梗死后不同时间静脉移植MSCs对心肌梗死大鼠心功能的影响,为MSCs移植的临床应用提供实验依据.
