| | 临床医学毕业论文【关键词】 BET-2细胞;电离辐射;线粒体跨膜电位(ΔΨm);细胞凋亡;过钒酸钠 摘要: 目的临床医学毕业论文 研究酪氨酸磷酸酶抑制剂过钒酸钠(Pervanadate,Per)对电离辐射作用后细胞线粒体膜电位(ΔΨm)改变和细胞凋亡的临床医学毕业论文影响。方法 应用流式细胞仪测定电离辐射作用后鼠源性髓系白血病细胞株BET-2细胞线粒体膜电位及其在过钒酸钠干预后的改变和细胞凋亡水平。结果 电离辐射早期可引起BET-2细胞线粒体膜电位降低,诱导细胞凋亡发生;过钒酸钠在一定程度上稳定细胞线粒体膜电位,减少细胞凋亡。结论 过钒酸钠可逆转电离辐射诱导的细胞线粒体膜电位的降低,减少造血细胞凋亡的发生。 关键词: BET-2细胞;电离辐射;线粒体跨膜电位(ΔΨm);细胞凋亡;过钒酸钠 Effects of pervanadate on MMP of BET-2 cells induced by ionizing irradiation Abstract: Objective To study the effects of pervanadate(Per),the inhibitor of protein tyrosine phosphotases on the alterations of mitochondrial membrane potential(MMP)and the apoptosis of BET-2 cells induced by ionizing irradiation.Methods BET-2 cells were divided into Per-treated groups and non-Per groups and incubated after ionizing irradiation.The MMP and apoptosis were measured with FCM.Results After irradiation was given,the MMP of BET-2 cells was reduced in the early stage significantly,and the apoptosis appeared,and these changes had a dose-dependent pattern and a time-course fashion.Per potently enhanced the MMP of BET-2 cells and prevented irradiation-treated cells from apoptosis.Conclusion Per reversed the repression of mitochondrial membrane potential and suppress apoptosis induced by ionizing irradiation in the hematopoietic cells. Key words: BET-2 cell;ionizing irradiation;mitochondrial membrane potential(MMP,ΔΨm);apoptosis;pervanadate(Per) 细胞凋亡是电离辐射导致造血系统损伤的主要方式。预防与控制造血干/祖细胞大量凋亡,有效恢复机体造血功能,在未来可能爆发的核事故、核战争以及肿瘤放射治疗副反应控制等领域都具有十分重要的意义〔1〕。造血细胞信号转导过程受蛋白质酪氨酸磷酸酶(protein tyrosine phosphotases,PTP)的调控〔2〕。过钒酸钠(pervanadate,Per)作为PTP特异性抑制剂,参与细胞内多条信号通路信息的传递过程的调节〔3〕。通过改变PTP活性调控造血细胞受体信号转导通路,可能成为继重组人粒细胞集落刺激因子(G-CSF)、促红细胞生成素(EPO)、重组血小板生长因子(TPO)等细胞因子之外的促进辐射造血损伤修复的发展方向〔4〕。本研究观察Per对照射后BET-2细胞线粒体膜电位改变及细胞凋亡的影响,探讨调控造血细胞受体信号通路中PTP活性对造血细胞电离辐射损伤的保护作用。结果报告如下。
