| | 中药炮制论文【关键词】 铅;硒;海马;神经元;神经突起 摘要: 目的中药炮制论文 观察铅、硒对原代培养的中药炮制论文海马神经元生长及存活的影响,研究硒对铅的神经细胞生长抑制的拮抗作用。方法 建立新生Wistar大鼠海马神经元原代无血清培养技术,通过PbCl2,Na2SeO3单独及联合作用,观察神经细胞的生长和存活情况。结果 10-5mol/L铅明显抑制了原代培养的海马神经元突起生长,引起神经元存活率下降;单独硒(2×10-6mol/L)有明显促进海马神经元突起生长的作用,尤其在神经元突起生长早期;但对海马神经元存活率的影响不明显。铅和硒共同孵育时,培养早期硒能拮抗铅对神经元突起延伸的抑制,但随着培养时间延长,这种拮抗作用消失;从神经元存活率来看,在实验剂量下,未见硒能拮抗铅对神经元存活的抑制作用。结论 铅具有抑制神经元生长和存活的毒性,本实验剂量下的硒在细胞培养早期有促进神经元突起生长和拮抗铅对神经元生长和存活的抑制作用。 关键词: 铅;硒;海马;神经元;神经突起 Effect of lead and selenium on primary cultured hippocampal neurons. Abstract: Objective To study the effect of lead and selenium on the growth and survival of primary cultured hippocampal neurons.Methods The primary culture of hippocampal neurons of newborn Wistar rats with serumfree medium was established.After cocultured with PbCl2 and Na2SeO3 respectively and simultaneously,the growth of neurites and survival of neurons were tested.Results Pb2+(10-5mol/L)delayed the growth of neurons,shortened the length of neurites,and decreased the neurons survival rate.Selenium(2×10-6mol/L) stimulated the growth of neurites,especially at early time of culture.There was no significant effect of selenium at studied dosage on survival rate of hippocampal neurons.The results of cocultivated with PbCl2 and along with the culture time prolongation,the antagonistic effect of selenium declined.The antagonistic effect of selenium on the inhibition of neurons survival rate caused by lead wasnt observed.Conclusion Lead inhibited the growth and survival of hippocampal neurons.At studied dosage selenium accelerated the growth of hippocampal neurons and antagonized the lead toxicity at the early culture time. Key words: lead;selenium;hippocampus;neuron;neurite 铅具有显著的神经发育毒性。毒理学实验和人群流行病学调查表明,铅能对神经系统造成不可逆性损害,严重损坏海马神经元,影响学习、记忆及行为发育〔1,2〕。硒有防止铅吸收和加速其排泄的作用,因此,硒有可能成为有效防治铅神经毒性的重要资源〔3〕。为了解硒拮抗铅神经毒性的效果和研究其拮抗作用机制,同时也为更多有效地利用硒,本试验对体外原代培养的Wistar乳鼠海马神经元进行低剂量PbCl2和Na2SeO3染毒,观察低水平铅、硒单独和联合作用对神经细胞生长和存活的影响。
