| | 超声医学论文【关键词】 乳腺癌;端粒酶;增殖细胞核抗原;多聚酶链式反应;免疫组织化学 摘要: 目的超声医学论文 探讨端粒酶活性与增殖细胞核抗原(PCNA)表达之间的超声医学论文关系及联合检测对乳腺癌术后治疗方案制定和预测预后的意义。方法 采用端粒重复序列扩增聚合酶链反应-酶联免疫吸附测定法(TRAP-PCR-ELISA)和免疫组织化学链霉素生物蛋白过氧化物酶(S-P)法对80例乳腺癌组织、36例乳腺癌相应癌旁组织和10例乳腺良性病变的端粒酶活性及增殖细胞核抗原进行检测。结果 (1)80例乳腺癌患者癌组织标本、36例乳腺癌相应癌旁组织标本中端粒酶表达阳性率分别为650%和83%,差异有统计学意义(P<001);10例乳腺良性病变组织标本中未检测出端粒酶活性;腋窝淋巴结有无转移与端粒酶阳性表达率差异无统计学意义(P>005)。(2)80例乳腺癌组织中PCNA的阳性表达率为6375%;乳腺癌有淋巴结转移组PCNA阳性表达率显著高于无淋巴结转移组(P<001),且呈正相关(P<001),即淋巴结转移程度越低,PCNA阳性表达率亦越低。(3)端粒酶活性与PCNA表达具有一致性(P<001),且呈正相关(P<001),一致率为8625%。结论 端粒酶活性与PCNA表达具有一致性,端粒酶活性与PCNA联合检测,可为乳腺癌术后治疗方案的制定和预测预后提供辅助依据。 关键词: 乳腺癌;端粒酶;增殖细胞核抗原;多聚酶链式反应;免疫组织化学 Relationship between telomerase activation and PCNA expression in breast cancer Abstract: Objective To approach the interrelation between telomerase activation and PCNA expression in breast cancer and evaluate the significance of combined detection in formulating therapeutic regimen after breast cancer operation.Methods The level of telomerase activation and PCNA expression were detected in 80 cases of breast cancer,36 cases of side-cancer tissues and 10 cases of lacteal gland benign lesion using TRAP-PCR-ELISA and S-P immunohistochemistry technique.Results (1)The positive rate of telomerase in 80 breast cancer tissues and 36 side-cancer tissues were 65.0% and 8.3% respectively and the difference revealed statistical significance(P<001),the telomerase activation was not examinated in 10 cases of lacteal gland benign lesion;It is no significance in statistic that the level of positive rate of telomerase and the extent of axillary nodes metastasis(P>005).(2)Among 80 cases ,the positive rate of PCNA in breast cancer tissues was 63.75% and the positive rate of PCNA in group with lymphatic metastasis was significantly higher than that in group without lymphatic metastasis(P<001) and there was positive relationship between lymphatic metastasis and positive expression of PCNA(P<001).(3)The telomerase activation was in accordance with PCNA expression(P<001) and there was positive relationship between telomerase activation and PCNA(P<001),the concordance rate was 86.25%.Conclusion The telomerase activation is in accordance with PCNA expression,associating telomerase activation with PCNA detection can provide the ancillary evidence to predict the prognosis and to formulate the treatment plan after breast cancer operation. Key words: breast cancer;telomerase;proliferating cell nuclear antigen(PCNA);polymerase chain reaction(PCR);immunohistochemistry 增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)是一种表达细胞增殖周期的36kd多肽,直接参与细胞增殖过程中DNA的复制,其表达程度能客观反映肿瘤细胞的增殖活性,过度表达增加了瘤细胞转移趋势的危险性,PCNA可作为判断肿瘤预后的指标〔1〕。为探索乳腺癌组织端粒酶活性与增殖细胞核抗原表达的关系,对80例乳腺癌组织、36例乳腺癌相应癌旁组织和10例乳腺良性病变的端粒酶活性及增殖细胞核抗原进行检测。
