【关键词】 脑缺血;再灌注;丝裂原激活蛋白激酶类;细胞凋亡;梗塞,大脑中动脉;大鼠 Expression of extracellular signalregulated kinase in focal cerebral ischemia 【Abstract】 AIM: To study the role of extracellular signalregulated kinase (ERK) during focal cerebral ischemiareperfusion in hippocampal neurons of rats. METHODS: Sixty male adult Wistar rats were randomly divided into 2 groups (n=30 per group): the sham operation group and the ischemiareperfusion group(I/R group). The rats of I/R group were subjected to 2 h of right middle cerebral artery occlusion (MCAO) with introducing a nylon suture to the right internal carotid artery and then 3, 6, 24, 48 and 72 h of reperfusion, respectively(as 5 subgroups, n=6 in each subgroup). The histopathologic changes were observed by HE staining. The apoptotic neurons were detected in CA1 area of hippocampus by TUNEL method. ERK phosphorylation was detected by immunohistochemistry. RESULTS: In I/R group as compared with shamoperation group, after MCAO, the survival neurons in CA1 areas decreased; a lot of apoptotic cells were observed and peaked at 48 h; ERK activity increased 3 h after MCAO and peaked at 6 h and returned to the normal level at 72 h. CONCLUSION: Cerebral ischemiareperfusion may result in the upregulation of ERK activity, and participate in the process of neuron apoptosis.
