| 试剂红细胞的研究与应用
| | 动物医学论文［摘要］目的动物医学论文：研制试剂红细胞用于血型鉴定反定型，保证临床输血安全。方法：制备效期长的动物医学论文红细胞保养液，再用该保养液制备4%的试剂红细胞，2℃～8℃冰箱保存，每周对其进行红细胞抗原性和免疫特异性测定及反定型试验，观察细胞凝集状况。结果：本法制备的试剂红细胞保存期为68 d，具有较好的抗原特异性和细胞稳定性，68 d以后凝集强度减弱。结论:该试剂红细胞凝集效能良好，红细胞抗原稳定性、免疫特异性好，具有成本低，制作易，操作简便等优点。

Study and Application of the Reagent Red Blood Cells

Abstract:Objective To study the reagent red blood cells which is used in the reverse typing of ABO insure the safety of clinical transfusion.Methods We make the nutrient liquor for keeping the red cell alive,and by using this nutrient liquor to produce the 4% the reagent rde blood cells that is kept under the 2 ℃～8 ℃ in the refrigerator.Every week we test the antigen, immunological specificity and dosome reverse typing experiments of the rde cells with the 4% the reagent red blood cells in order to observe the condensation of cells.Results The reagent red blood cells can exist sixtyeight days,by this method we have the better antigen specificity and stability of cells.After sixtyeight days the intensity of condensation will be brought down.The condensation of this reagent red blood cells is fine,the immunological specificity and the stable antigen of the red cells is good.At the same time this method have many advantages, such as the easy method and operation, the cheaper expense and so on.

临床输血用于血型反定型的试剂红细胞购置较困难，且保存期、运输等问题常常影响工作，自制的试剂红细胞效期短，红细胞抗原稳定性和免疫特异性及凝集效能不能保证，导致血型鉴定错误，甚至出现溶血性输血反应。笔者探索了一种效期长的红细胞保养液，并制备出试剂红细胞，2 ℃~8 ℃保存期达68 d，具有良好的细胞稳定性和免疫特异性。
1 材料与方法
1.1 红细胞保养液 枸橼酸0.66 g、枸椽酸钠5.48 g、磷酸二氢钠2.22 g、腺嘌呤0.26 g、肌苷0.22 g、甘露醇7.5 g、葡萄糖19.9 g、氯化钠8.35 g、EDTA2NA 120 mg、叠氮钠40 mg、防腐防霉剂400 mg、稳定剂适量、水加至1 000 ml，用3%碳酸钠调至pH 7.2～7.4,高压灭菌。
1.2 仪器 净化工作台、离心机。
1.3 试剂 单克隆抗A、抗B血型定型试剂， 由长春博德生物工程有限公司提供，批号：20060223,质量控制符合要求，在有效期内使用。
1.4 人源抗A、抗B、抗AB血清 均由3个同型人全血等到量混合,分离血清而成,无不规则抗体。
