人类β1,3半乳糖基转移酶7对人4IgB7H3的修饰作用初探

【执业药师论文】作者:唐春花, 马震宇, 岳爱环, 行书丽,蒋俊华 张昊 曹静媛, 周迎会,吴士良【摘要】 目的: 探讨人4IgB7H3基因转染人胃癌细胞株SGC7901后人类β1,3半乳糖基转移酶7(β1,3galactosyltransferase 7,β3GalT7)的表达变化,以期发现4IgB7H3与β3GalT7的相互关系。方法: 脂质体介导重组逆转录病毒载体pEGZTerm/4IgB7H3转染SGC7901细胞,筛选获得稳定表达4IgB7H3的亚克隆细胞株,通过RTPCR和蛋白质印迹方法检测β3GalT7的表达。结果: 成功获得稳定表达4IgB7H3的亚克隆细胞株SGC7901/4IgB7H3;β3GalT7随4IgB7H3的表达上调而上调。结论: 4IgB7H3蛋白的修饰成熟可能需要β3GalT7的作用。 【关键词】 4IgB7H3; β1,3半乳糖基转移酶7; SGC7901; 转染 [Abstract] Objective: We researched the change of UDPGal:betaGlcNAc beta 1,3galactosyltransferase polypeptide 7 expression in SGC7901/4IgB7H3 cell to find the relation between 4IgB7H3 and β3GalT7. Methods: Transfered pEGZTerm/4IgB7H3 into SGC7901 cell, then detected the expression of UDPGal:betaGlcNAc beta 1,3galactosyltransferase polypeptide 7 with RTPCR and Westernblotting.
