高效液相色谱法测定栽培射干中次野鸢尾黄素的含量

【##西医 论文】【摘要】 目的 应用 高效液相色谱法测定栽培射干中次野鸢尾黄素的含量,考察其药材的内在质量。 方法 采用高效液相色谱法,色谱柱为TSK ge1 ODS 80 TS250 mm×4.6 mm;流动相为甲醇磷酸水溶液(pH=3)∶53%∶47%;流速为1 ml/min;检测波长为266 nm;柱温35℃。结果次野鸢尾黄素在0.0118~0.118 μg范围内与色谱峰面积呈线性关系(r=0.999 9);平均加样回收率为99.84%,RSD为2.059%,次野鸢尾黄素的含量为0.1252%。结论该法简便、准确、灵敏度高,栽培射干中的次野鸢尾黄素的含量大于0.10%,符合《 中国 药典》要求。 【关键词】 高效液相色谱法 射干 次野鸢尾黄素 Abstract:ObjectiveTo determine irisflorentin in cultivated Belamcanda chinensis(L.)DC by HPLC,and to inspect the quality of its ingredients.MethodsUsing HPLC with TSK ge1 ODS Column80(250 mm×4.6 mm),the mobile phase was composed of a mixture of methanol-phosphoric aicd aqueous soluton (pH=3)∶53%∶47%,the flow rate was 1 ml/min,the UV detection wavelength was set at 265 nm,and the temperature was at 35℃.ResultsIrisflorentin showed linear relationship with peak areas in the range of 0.0118~0.118 μg(r=0.999 9). The average recovery was 99.84%,RSD was 2.
